Description
- Introduce of Flp Recombination Target (FRT) site into genome of mammalian cell line of choice
- Expression vector containing gene of interest is then integrated into genome via Flp recombinase-mediated DNA recombination at FRT site
- Major components include Flp-In. target site vector, pFRT/lacZeo, for generation of host cell line containing integrated FRT site
- Expression plasmid containing a FRT site linked to hygromycin resistance gene for Flp recombinase-mediated integration and selection of stable cell line expressing gene of interest under control of human cytomegalovirus (CMV) immediate-early enhancer/promoter
- Flp recombinase expression plasmid, pOG44, for expression of Flp recombinase under control of human CMV promoter
- Control expression plasmid containing chloramphenicol acetyl transferase (CAT) gene, which when cotransfected with pOG44 into the Flp-In. host cell line, expresses CAT
