Description
The Sphericalplate 5D (SP5D) is a 3D cell culture plate for the formation of uniform, standardized, and size-controlled spheroids in high quality and yield. It also allows you to effortlessly scale up and further translate into diagnostic or clinical applications. Simultaneously, it increases the reproducibility of subsequent tests, as you always start your experiments with identical initial conditions due to the low size variance of the cultured cell spheroids.
The patented geometry with special coating allows the integration of every single cell into a 3D-agglomerate in a controlled way. This is how uniformly sized spheroids grow without the formation of necrotic cores.
Design:
- Patented, three-dimensional geometry with rounded bottoms of the microwells, resulting in scaffold-free, regular spheroid formation while keeping the aggregate in the center.
- The microwell geometry prevents hypoxic damage through aggregate size control.
Ease of Use:
- There are only three main steps in the SOP and it just takes 24 hours for your 9,000 spheroids to be produced.
- Step 1) Prepare functional well with medium. Step 2) Add single cell suspension. Step 3) Incubate.
- Plates are sterilized and have pre-applied anti-adherence coating, making them ready to use out of the box. No pretreatment or centrifugation steps after seeding your cells.
- Patented surface coating to prevent cell-surface signaling during development and eliminates the need for plate pre-treatment.
- Special angle of the microwells to allow cells descending in consistent numbers without an additional centrifugation step.
Performance:
- Spheroid size is controlled by initial seeding density.
- No sharp edges at the bottom of the plate to reduce surface-induced, non-physiological differentiation.
- Sharp edges at the top of the microwells to leave no cell behind and no unwanted cell growth outside of the microcavities.
- Robust non-fouling surface coating featuring low protein-adhesion to allows perfect cell aggregation and prevents surface adhesion.
- Aggregate size control through the geometric dimensions of the microwells to avoid hypoxia.
- Selection of COC (cyclic olefin copolymers) material to enable optimal real-time imaging with less background noise.
Applications:
- Standardization of geometric cluster size for clinical applications.
- A wide range of cell lines, islet cells, and stem cells have been used on this plate successfully.
24-Well Plates:
- Standard 24 circular well plate dimension (12 wells have microwells (3D spheroid production), 12 wells do not have microwells (control)).
- Each well has 750 microwells, allowing the plate to produce up to 9,000 spheroids.
- Comfortably carry out monolayer comparative studies in the remaining 12 wells of the same plate.
6-Well Plates:
- Standard 6 square well plate.
- Expected yield of 3,364 spheroids per well, thus a total yield of 20,184 spheroids per plate.
- Suitable for scale-up trails and bigger batches.
