Dolichos biflorus agglutinin (DBA) binds α-linked N-acetylgalactosamine. A glycoprotein consisting of four subunits of approximately equal size and with a molecular weight of about 111 kDa, DBA is used to establish secretor status in blood group A individuals by hemagglutination inhibition techniques and for blood typing. It has also been used as a general marker of developing renal collecting ducts. This fluorescein-labeled DBA features a ratio of fluorophores to lectin protein that provides optimal staining (excitation 495 nm, emission 515 nm). Supplied as a solution essentially free of unconjugated fluorophores, it is preserved with sodium azide. The recommended inhibiting/eluting sugar is 200 mM N-acetylgalactosamine.

The treatment of PVT-WGA SPA beads with positively charged polyethyleneimine (PEI) blocks potential non-specific binding sites on the SPA bead surface. There are two SPA bead types available with PEI treatment. The PVT-WGA-PEI type A SPA beads are treated with PEI prior to the coupling of WGA to the PVT SPA bead. The PVT-WGA PEI type B SPA beads are treated with PEI after the WGA coupling stage. The PVT-WGA-PEI type A and type B SPA beads exhibit different characteristics with regard to the non-specific binding of radiolabelled ligand directly to the SPA bead. Therefore, both bead types should be evaluated when deciding which SPA bead to use and both are included in the Select-a-Bead Kit. The binding capacity of both bead types for cell membrane protein remains 1030 g membrane protein per milligram of SPA bead. Sambucus Nigra Lectin (SNA, EBL), CY3, 1 mg, isolated from elderberry bark, binds preferentially to sialic acid attached to terminal galactose in alpha-2,6 and, to a lesser degree, alpha-2,3 linkage. Binding is also inhibited to some extent by lactose or galactose. This lectin does not appear to bind sialic acid linked to N-acetylgalactosamine. SNA has been reported to inhibit cell-free protein synthesis. Cy3-labeled Sambucus nigra lectin has an appropriate number of fluorochromes bound to provide the optimum staining characteristics for this lectin.
